If I have a protein sample which after plotting a calibration curve I've determined to be say 20 ug/mL in concentration, how do you adjust such sample to a particular concentration e.g. 15 ug/mL? By simple proportion by adding distilled water or buffer?

You could dilute it with distilled water.
