Why doesn't changing the enzyme concentration change the Km? If you go from having 10000 enzymes to having only 2 enzymes, then wouldn't you need a smaller substrate concentration (and therefore a smaller Km) to reach Vmax/2?

are you asking why it doesn't change Km for competitive vs noncompetitive inhibitors? I think this is what you're referring to.

if not then no, you wouldn't need a smaller substrate concentration to saturate 10000 enzymes vs 2..remember the free ligand approximation and the three assumptions that you have to make when dealing with michaelis-menten kinetics....if you have a lot of substrate, no adjustments to the concentration of the substrate needs to be made to saturate the enzyme.
