Mediator as a bridge between gene-specific regulatory factors and the general transcription

Description
a. Activation. Mediator has three distinct structural domains—head, tail, and middle. The tail domain interacts with transcriptional activators (ACT, in red) and links Mediator with RNA polymerase.

b. Repression. The four-subunit complex containing Srb8, Srb9, Srb10, and Srb11, when bound to Mediator, prevents its interaction with RNA polymerase II and the basal transcription machinery.
