While virtually any DNA sequence can be cloned into plasmids, one of the practical limitations is that:
 a. Only bacterial DNA can be cloned into bacterial plasmids.
 b. Only very large pieces of DNA can be inserted efficiently into plasmids.
 c. Only plasmids with foreign inserts shorter than about 10 Kbp are replicated.
 d. Only plasmids with EcoR1 cloning sites provide a means for efficient expression of the foreign gene.
 e. Inserts of foreign genes must always have exactly the same restriction site on both ends.
