I couldn't find much on the web to help me out...

Predict the effect on GAL1 transcription, in the presence of galactose alone, of the following mutations (An example of the prediction of the effect for the mutation in the question_a was provided):

Mutants   Predict the effect

a) Deletion of all four Gal4-binding sites in the GAL1 UAS element   

The deletion of all four Gal4-binding sites from the UAS would prevent transcriptional activation of the GAL1 gene.

b) Deletion of one Gal4-binding site in the GAL1 UAS element   

c) Deletion of the Gal4 activation domain   

d) Deletion of the GAL80 gene   

f) Deletion of the GAL1 promoter   

g) Deletion of the GAL3 gene   


I'm gonna keep reading the book to hopefully understand this.  Thanks.

Anyone understand this problem? Think you can explain to me how to do it?

b. gene activation will not occur in vivo, and thus the genes are silent.

c. transcription will not be activated, so the lac z gene will remain off.

d. GAL gene expression will not be inhibited, and Gal4 protein will not be regulated, and thus GAL structural genes will be transcribed, when galactose is not present.

f. Neutral Face?

g. structural genes will not be active in presence of galactose.

That's all I've found so far, not sure if any of it is correct :/
