Ans: DNA polymerases require a free 3-OH end of either RNA or DNA, called a primer, in order to initiate DNA synthesis. In DNA replication, this is most often accomplished by the synthesis of a short RNA sequence from the DNA template by a primase, an RNA polymerase that does not require a free 3-OH end. Primers can also be generated by the hybridization of an already synthesized RNA to the template strand, or by the nicking of a strand of the DNA. In some cases, a protein can directly prime the polymerization reaction.
