 Because eukaryotic RNA is processed to form mRNA, it is important to use eukaryotic mRNA to make cDNA before the genes are inserted into a bacterial host. The eukaryotic mRNA can be extracted using chromatography followed by the use of a low salt buffer to remove the mRNA from the chromatography column. RT-PCR is used to make cDNA, which is inserted into an expression vector.
