What factors would help determine the insertion of a gene into a vector? I'm trying to find which enzyme is better for inserting a recA gene into a pBC KS+  vector? ClaI or EcorI?

1)  If recA has an internal site for either of these restriction enzymes, you wouldn't want to use that one.

2)  Same thing for the vector itself, except that you *do* want the vector to have one or two sites for either Cla I or EcoRI (so you can get the recA gene in).
