the lectin  receptor was solubilized by treating cell membranes with non-ionic detergent Triton X-100. What single procedure could I do to isolate the lectin receptor? Is it along the lines of purifying it and using a column?

Use the lectin it binds.
If you attach its lectin ligant to a sephadex column, and pass the mixture down it, then (theoretically) only your lectin receptor will bind it.

You can then wash it off with elevated salt.
