Theoretically, all you'd need is a single sample. All the DNA you need to clone a human being is from a single nucleated cell.

Is this in the direction in which you were expecting the question to be answered?

I mean if one simply had a DNA readout on computer. Could all differences that would make that human unviable be detected?

Depends on your definition of viable. Assuming that by viable you mean one who possesses all the genes that make up a human being, then technically yes. You could technically  identify whether the DNA sample in question possesses any of the genes you'd normally find in a true DNA sample from a human being.

You could confirm that the dna could create a living human that wouldn't turn into a giant tumor or otherwise terminate before birth?

Theoretically, any true sample of human DNA possesses all the information needed to recreate that person. Development of humans, however, isn't fully understood, so chances are you will end up with a giant tumor or some dysfunctional blob of tissue.
