I know that heat, pH, concentration of enzyme and substrate play an important role in enzyme conformation and ultimately in the activity of the enzyme. But is there any other factors that also contribute to the maximum activity of the enzyme? How do the experimenters know that a specific structure of the enzyme correlates to the enzyme activity?

Competitive and non-competitive inhibitors will also affect the enzyme activity. With competitive inhibition, if the substrate concentration is increased sufficiently, the enzyme will reach its maximum efficiency eventually. With non-competitive inhibition, increasing the substrate concentration will not allow the enzymes to reach maximum activity.
