the protease inhibitor doesn't break open the cells it's there to make sure the proteins on the inside aren't broken down when the cell lyses (the proteases are released when the cell lyses)

the time(s) we used it, we used a protease inhibitor cocktail, it has all sorts of inhibitors to block most/all proteases.
