the protease inhibitor doesn't break open the cells it's there to make sure the proteins on the inside aren't broken down when the cell lyses (the proteases are released when the cell lyses)

the time(s) we used it, we used a protease inhibitor cocktail, it has all sorts of inhibitors to block most/all proteases.

http://www.sigmaaldrich.com/life-science/proteomics/recombinant-protein-expression/cell-lysis/protease-inhibitor-cocktails.html check out the datasheet, there's also a special one for yeast (not meant as spam i just googled protease inhibitor cocktail to get an example)
