I have a molecular biology question, can you help me to solve it?

You have cloned a cDNA molecule in the EcoRI and PstI sites of pBR322. Which bacterial strain (XLI-blue or JM105) are you going to use for the transformation experiment and briefly explain why?
(E. Coli  genotypes)
Xl1-blue lac- F’ [ lacIq lacZΔM15 Tn10(Tetr)]
JM105 lac- F’ [ proAB+ lacIq lacZΔM15]

thank you very much.

This one is better because it is partly restriction-deficient and an overall good strain for cloning repetitive DNA.

CTTCTCGCCATTG                       CGCTCAATTGCGC
GAAGAGCGGTAAC                    GCGAGTTAACGCG

Design two primers to clone this DNA fragment
in the EcoR1 site of pUC18

                             AND
Design two primers to clone this DNA fragment
in the EcoRI and HindIII sites of pUC18

EcoRI = GAATTC      HindIII = AAGCTT

thanks again.
