Dot blot is a technique for detecting and identifying proteins, similar to the Western blot technique but differing in that protein samples are not separated electrophoretically but are spotted through circular templates directly onto the membrane or paper substrate. Antigens may be applied directly to nitocellulose membrane as a discrete spot (dot) to give a simple and reliable assay.

An overlay assay is a biological technique used to find proteins that bind to a protein of interest. It begins with proteins that are dispersed but fixed (e.g. a tissue slice or run on a membrane by gel electrophoresis) and then incubated in the labelled protein of interest. The label will indicate the location of proteins that bind to the protein of interest.

Two-hybrid screening or 2-hybrid screening is a molecular biology technique used to discover protein-protein interactions and protein-DNA interactions by testing for physical interactions (such as binding) between two proteins or a single protein and a DNA molecule, respectively.

The premise behind the test is the activation of downstream reporter gene(s) by the binding of a transcription factor onto an upstream activating sequence (UAS). For the purposes of two-hybrid screening, the transcription factor is split into two separate fragments, called the binding domain (BD) and activating domain (AD). The BD is the domain responsible for binding to the UAS and the AD is the domain responsible for activation of transcription.
