I have to study the impact of temperature on bacteria. So I cultured Vibrio cholerae in a broth. I calculated the number of bacteria in broth by using serial dilution technique. After that, I transferred 1ml of the bacteria from broth into sterile seawater to be exposed to the heat. After done with the heat treatment, I wanna know what is the number of bacteria that still alive in that sterile seawater. What method should i use? Is it the serial dilution too? Or can I use membrane filtration? Since I have to compare the number of the bacteria before and after the heat treatment. Thanks a lot
