What are the steps in the shotgun approach to whole-genome sequencing?

Step 1)             DNA isolation from the cell

Step 2)             Shearing whole genome into fragments by ultra-sonication or restriction enzymes

Step 3)             Selection of average sized fragments (approximately 2kb)

Step 4)             Ligation of fragments into appropriate vectors

Step 5)             Vectors are transformed into E.coli for amplification of desired fragments.

Step 6)             Desired fragments are sequenced by using chain termination method

Step 7)             Assembling of sequenced fragments into one full length using computer algorithm
