Hello

I believe the most basic way is to first isolate the DNA using any known technique such as phenol-chloroform extraction (linked below). Once the DNA of the species has been isolated, the DNA is stained and the chromosomes can be visualized under a microscope using staining techniques, such as Giemsa staining or fluorescent dyes that bind to DNA. During cell division (specifically, metaphase), chromosomes condense and become visible.
