I was wondering if ELISA could detect nucleic acids and If so why? and if not, why not?

i would have to say no... The ELISA format by definition is used to detect antigens with antibodies. So since there's no antibodies for DNA or RNA, you cannot do ELISA. Perhaps a similar method is chip on chip, where you immunoprecipitate the DNA with antibodies detecting transcription factors or activators etc. And then bind those DNA to a microarray
