These problems aren't making sense, please help.

You do a conjugation experiment in which you mate Hfr leu- , strr , ampr , purr, and F- leu+, strs, amps, purs. In all cases resistance is dominant. Strr is closest to the fertility factor. When plated on minimal media containing the supplements listed below you obtain the following data:

strr ampr purr 500 colonies
strr ampr purs 0 colonies
strr amps purr 200 colonies
strr amps purs 10 colonies

If a map unit is equal to 1% recombination then what are the map distances?



You are mapping the bacterial genome of a strain of E. coli. You set up the following cross. Hfr ampr, his+, pro+, purs, thr+, trp- and F- amps, his-, pro-, purr, thr-, trp+. Thr is closes to the fertility factor. You plate these cells on minimal media with supplements. After one hour you interrupt the mating and obtain colonies with the following genotypes.

amps, his+, pro+, purs, thr-, trp- 647 colonies
ampr, his+, pro+, purs, thr-, trp- 102 colonies
amps, his-, pro+, purr, thr-, trp+ 1000 colonies
ampr, his+, pro+, purs, thr+, trp- 0 colonies
amps, his+, pro+, purr, thr-,trp+ 875 colonies
amps, his+, pro+, purr, thr-, trp- 802 colonies

What is the gene order and how did you determine this order?

Hey bio_man yep, this is just your basic microbial genetics i figured it out! Basically for the first one you look at your 0 colony offspring to determine gene order and you turn your recipient ( F- ) and your donor ( Hfr ) into your offspring. Once you get them figured out you put those two over the total and times by 100. For the second one you look at the Hfr ampr his+ pro+ purs thr+ and trp- and see if they changed, like for the 1000 colonies you can see that pro+ changed because the others are all opposite and pro+ is still pro+ therefore it changed and you do that for all of them but you ignore pro+ because you already did it and you ignore the ones you already used until you get down to the last one and you have your order.
