A plate containing several different colonies can be probed for a particular sequence of interest using colony hybridization. After replica plating (to save viable cells), one replica set of colonies are lysed and their nucleic acids are bound onto a filter paper. This paper is then subjected to a traditional Southern blot using a specific nucleic acid probe to observe the presence of a particular gene of interest.
